Abstract:
Light sheet microscopy or selective plane illumination microscopy (SPIM) has proven to be a powerful imaging technique due to its unsurpassed acquisition speed and gentle optical sectioning. This allows for in vivo recordings of entire biological specimens, bridging the spatio-temporal scales between fast subcellular processes and organismal dynamics. In this lecture I aim to give an overview of this highly dynamic field of microscopy, from early developments to recent advances, emphasizing multi-view techniques that allows for in toto imaging of small model organisms e.g. the fruit fly embryo. Finally, I will discuss a couple of promising future applications of light sheet based fluorescence microscopy.
